How is the tumor cytotoxicity assay conducted?
After thawing, NK cells are cultured in NK cell cryopreservation medium for 4 hours and 12 hours, then co-incubated with target cells for 4 hours. Tumor cell death is evaluated using flow cytometry.

Has physiological saline been used to dilute NK cells post-thawing to achieve a specific cell density for viability stability testing in the NK cell culture protocol?
No, but this can be arranged upon request.

Does the NK cell cryopreservation solution include DMSO, and if so, what is its approximate concentration?
Both NK1 and NK3 cryopreservation solutions contain approximately 10% DMSO.

Have you tested the viability stability of NK cells in cryobags using the NK cell culture protocol?
Not yet. All tests have been conducted in cryovials. We have evaluated NK cell stability in cryovials for up to 6 hours in cryopreservation solution before freezing and after thawing. For T cells, extensive cryobag testing has been performed.

Why do NK cells, cryopreserved for 2 weeks and cultured for 48 hours in NK cell culture media, show clumping and low viability (30%) despite 87% pre-freeze and 82.7% post-thaw viability? Is handling a factor?
NK cells naturally have low post-thaw viability, with clumping indicating dead cells. The NK cell source and medium are key factors. Optimizing the medium with human albumin helps. Cryopreservation solution (even CS10) has minimal impact. See our PPT for client data.

A client reports that the NK3 cryopreservation solution is viscous and forms bubbles during use in the NK cell culture protocol. How can this be prevented?
For freezing, use reverse pipetting (draw at second stop, dispense at first stop). Mix gently, avoid fully expelling the solution during pipetting, keep liquid in the tip, and release along the tube wall to avoid injecting directly into the suspension.

Do you have data on NK cells cultured for 24 hours post-thaw in NK cell culture media?
Yes, our brochures and PPT include post-thaw culture data at 12, 24, 36, and 48 hours.

Is there high-density cryopreservation data for NK cells?
Internal tests show viability at 2E8 and 5E8, but we recommend ≤2E8 for clients. Some clients successfully use 2E8.

What is the longest duration for which NK cell cryopreservation data is available?
Data are available for up to 12 months.

Has the viability of NK cells been tested after 1 month at -80°C in NK cell medium?
Yes.

Does Cellstore currently offer DMSO-free cryopreservation solutions for NK cells in the NK cell culture protocol?
Not yet. Current DMSO-free prototypes achieve >90% recovery but lack sufficient tumor cytotoxicity.
